Headspace solid-phase microextraction combined with gas chromatography/ion trap tandem mass spectrometry (HS-SPME/GC/ITMS/MS) was used for the analysis of 12 halobenzenes from soil samples. For MS/MS optimisation, the experiments were performed by precursor ion selection and software controlled operations. Collision-induced dissociation (CID) can be achieved by two different approaches, resonant and non-resonant excitation modes. Different results were obtained using the two approaches, and the resonant excitation mode was chosen as the best for all halobenzenes. Parameters such as the CID excitation amplitude, excitation RF storage level and CID bandwidth frequency were optimised to maximise the formation of halobenzene product ions. A 100-microm polydimethylsiloxane fibre was used for the isolation and preconcentration of the analytes. The HS-SPME/GC/ITMS/MS method was applied to the analysis of halobenzenes in an agricultural soil sample. The halobenzenes were quantified by standard addition, which led to good reproducibility (RSD between 4.7 and 9.2%) and detection limits in the low pg/g range. The method was validated by comparing the results with those obtained in a European inter-laboratory exercise.  相似文献

This study describes the application of capillary electrophoresis (CE) to the analysis of ebrotidine and its metabolites as an alternative analytical technique to liquid chromatography. Comparison between UV-diode array spectroscopy and mass spectrometry (MS) using an ion-trap system with electrospray ionization as detection systems has been performed. The quality parameters of the UV detection method were established, obtaining linear calibration curves over the range studied (8-200 mg ml(-1)), limits of detection between 3.4 and 4.3 microg ml(-1), and run-to-run and day-to-day precision lower than 14%. For these compounds the protonated species [M+H]+ and, in some cases, sodium adducts were observed in the MS spectra. Using MS coupled to CE, limits of detection were between 0.5 and 2.6 microg ml(-1).  相似文献

Polychlorinated n-alkanes (PCAs), also known as chlorinated paraffins (CPs), are highly complex technical mixtures that contain a huge number of structural isomers, theoretically more than 10,000 diastereomers and enantiomers. As a consequence of their persistence, tendency to bioaccumulation, and widespread and unrestricted use, PCAs have been found in aquatic and terrestrial food webs, even in rural and remote areas. Recently, these compounds have been included in regulatory programs of several international organizations, including the US Environmental Protection Agency and the European Union. Consequently, there is a growing demand for reliable methods with which to analyze PCAs in environmental samples. Here, we review current trends and recent developments in the analysis of PCAs in environmental samples such as air, water, sediment, and biota. Practical aspects of sample preparation, chromatographic separation, and detection are covered, with special emphasis placed on analysis of PCAs using gas chromatography–mass spectrometry. The advantages and limitations of these techniques as well as recent improvements in quantification procedures are discussed.  相似文献

Absence of gradients and Nernstian equilibrium stripping (AGNES) is a recently developed electroanalytical technique specifically designed for the direct determination of free concentrations of metal ions. AGNES is applied here to the determination of free Zn concentration in a river water sample. The method has been validated with synthetic solutions of low ionic strengths containing Zn and 2,6-pyridinedicarboxylic acid and then applied to synthetic river waters and to a natural sample collected from Besòs River in Montcada i Reixac (Catalonia, North-Eastern Spain). In the river sample, an average free Zn concentration of 12.8(4) nM was obtained, while the total dissolved Zn concentration was 0.51(8)?μM. To control and maintain pH and pCO2 constant during AGNES measurements, a novel device for N2/CO2 mixed purging has been developed.  相似文献

This paper presents the development of a fast multi-residue method for the determination of 49 pharmaceuticals and 6 metabolites from different therapeutic classes in water resources by means of Ultra-performance™ liquid chromatography (UPLC) coupled to tandem mass spectrometry. The use of the UPLC technology enabled all the 55 compounds to be separated chromatographically in less than 9 min (6.3 min positive mode and 2.7 min negative mode) and with a total analysis time of 18 min when considering column conditioning. Improved resolution, sensitivity and a reduction of matrix effects were obtained under these conditions. Unequivocal identification and quantification of the target compounds was also performed by using the dual acquisition modes of the hybrid triple quadrupole-linear ion trap (QqLIT) system. Triple quadrupole mode by means of selected reaction monitoring (SRM) was used for quantification, whilst a second SRM transition together with information-dependent analysis (IDA) experiments was used for confirmation. Additionally, one general, single solid-phase extraction (SPE) method was developed by using Oasis HLB cartridges. Quality parameters of the method in wastewaters were established obtaining a fast, robust, reproducible and cost-effective method for all the target pharmaceuticals. Finally, the optimized SPE-UPLC/QqLIT method was used for the analysis of the target compounds in wastewaters from Spain. Thirty-one out of fifty-five compounds were identified in the samples collected.  相似文献

A one-step extraction and clean-up method using pressurized liquid extraction (PLE) (selective PLE) combined with gas chromatography-ion-trap tandem mass spectrometry (GC-ITMS-MS) was evaluated for the analysis of polybrominated diphenyl ethers (from tri- to hepta-PBDEs) at low concentrations in fish and shellfish samples. To this end, the performance of an on-line PLE extraction/clean-up method and of a classical Soxhlet extraction and clean-up method using a multi-layer modified silica column were compared. The two sample treatment methods provided similar results, although an important reduction in the sample treatment time (40 min per sample) was achieved using the selective PLE method. In addition, the suitability of the PLE combined with GC-ITMS-MS method was evaluated by comparing the results obtained in the analysis of fish samples with those obtained by gas chromatography-high resolution mass spectrometry (GC-HRMS). Good agreement between both techniques was obtained with differences between the mean values of less than 16%. The selective PLE method coupled to GC-ITMS-MS produced accurate results for PBDE determination with low limits of detection (1.0-16.8 pg g−1 wet weight) and quantification (3.1-51 pg g−1 wet weight) as well as good precision (RSD < 16%). This method has been applied to the analysis of PBDEs in fish and shellfish samples collected at fish markets in Catalonia (NE Spain).  相似文献

We present a fast liquid chromatography–tandem mass spectrometry (LC–MS/MS) method for the analysis of the coccidiostat amprolium in food samples. Tandem mass spectrometry in a triple quadrupole was used for quantitative purposes, and the information from multiple-stage mass spectrometry in an ion-trap mass analyzer contributed to fragmentation studies. Hydrophilic interaction liquid chromatography (HILIC) in a Fused-Core™ column using isocratic elution (acetonitrile:formic acid/ammonium formate buffer pH 4, 50 mM (60:40)) successfully analyzed this compound in less than 3 min. The HILIC system was coupled to heated electrospray-MS/MS using highly selective-selected reaction monitoring (H-SRM) to improve sensitivity and selectivity for the analysis of amprolium, after a simple sample treatment based on an “extract and shoot” strategy. Accurate mass measurements were performed to identify the interfering compound responsible for causing matrix ion enhancement in the signal of amprolium. The addition of l-carnitine (the interfering compound) (1 μg L−1) to standards and sample extracts allowed the use of the external calibration method for quantitative purposes. The LC–MS/MS (H-SRM) method showed good precision (relative standard deviation, RSD, lower than 13%), accuracy and linearity and allowed the determination of amprolium down to the ppb level (LODs between 0.1 and 0.6 μg kg−1).  相似文献

Gravitational field-flow fractionation (GrFFF) is applied to the fractionation of active dry wine yeast. An experimental approach to the analysis of the effects that field variation by changing mobile phase composition and flow-rate have on the fractionation process of standard particles (polystyrene) was first developed to further obtain effective fractionation of wine yeast by GrFFF. Scanning electron microscopy and Coulter counter particle size measurements were used to monitor the fractionation extent and capabilities of GrFFF to describe the distribution of yeast cells populations.  相似文献

Isopropylthioxanthone (ITX), usually applied as a mixture of 2- and 4-isomers, is a common photo-initiator in UV inks used in paper- or plastic-based packaging materials. In this work a pentafluorophenylpropyl column (HS F5) has been used to achieve the chromatographic separation of the two isomers. A gradient elution with acetonitrile and a 25mM formic acid-ammonium formate at pH 3.75 are required to provide an Rs of 1.3 between the two compounds. The fragmentation pattern of ITX was studied using two mass analyzers, an ion trap (IT) (multi-stage fragmentation) and a triple quadrupole mass analyzer of hyperbolic rods (accurate mass (AM) measurement). The protonated molecule [M+H](+) observed in the mass spectrometry (MS) spectrum lost an isopropyl group, [M+H-C(3)H(6)](+). Later, this ion fragmented, yielding the radical ion [M+H-C(3)H(6)-CHO](+). The elemental composition of these product ions was confirmed by AM measurement. Electrospray ionization (ESI) was used as an ionization source to couple liquid chromatography (LC) to MS. Instrumental quality parameters of three acquisition modes provided by the triple quadrupole mass analyzer were studied and good run-to-run precision (relative standard deviation, RSD, lower than 10%) and limits of detection (LODs) down to 0.8pg injected in the LC-MS/MS system were obtained. Finally the LC-MS/MS method using H-SRM Q1 acquisition mode was used to analyze 2- and 4-ITX in a range of food samples. The use of highly selective selected reaction monitoring (H-SRM on Q1) resulted in improved selectivity without sensitivity loss.  相似文献
